Unions are due to meet Royal Mail representatives to discuss the closure of the Gloucester depot in 2008.
Royal Mail wants to spend £20m replacing outdated buildings and introducing new technology.
Under the plans the mail centres at Reading and Gloucester will move to the expanded site at Rowland Hill Close, Dorcan, Swindon.
Unions said the move will add an extra two million road miles to deliveries, a claim dismissed by Royal Mail.
